品系详情
此品系为 Rag1tm1Mom 突变纯合小鼠,不会产生成熟 T 细胞或成熟 B 细胞。Prkdcscid 纯合小鼠可以产生一些 B 细胞和 IgM(即,“渗漏”),Rag1tm1Mom 纯合小鼠则缺乏所有类型的成熟淋巴细胞(即,“无渗漏”)。Rag1 敲除小鼠没有 CD3+ 或 T 细胞受体 (T cell receptor, TCR) αβ阳性细胞。杂合子或野生型同窝小鼠胸腺中细胞含量是突变小鼠的 15 到 130 倍。胸腺细胞为 CD8阴性CD4阴性 且大多为 IL2受体阳性。脾脏及骨髓中均不含 IgM 或 IgD 染色阳性的细胞,指示成熟 B 细胞完全缺失。这些数据和其他数据表明,B 细胞和 T 细胞发育在早期已经被阻断。通过肉眼观察难以区分该突变体与杂合子或正常野生型同胞小鼠。
品系建立
Rag1tm1Mom 突变品系由麻省理工学院癌症研究中心 Susumu Tonegawa 博士实验室的 Peter Mombaerts 博士建立。在品系建立过程中,使用了带 Pgk-neo 标记的靶向载体。该靶向载体引起的同源重组导致编码序列 5' 端有 1356 bp 长度的缺失。使用的是源自 129S7/SvEvBrd-Hprt+ 的 AB1 胚胎干细胞系。 将携带 Rag1tm1Mom 突变的小鼠与 C57BL/6J 近交小鼠回交十代,即获得 背景为C57BL/6J的 品系。
